Primary outcome
The main study endpoints are Hb trajectories and DNA sequence.
Secondary outcome
Secondary study parameters are iron parameters, iron deficiency-related
symptoms, and Hb deferral.
Background summary
Prediction of Hemoglobin (Hb) deferral may be helpful in the management of the
donation program. Recently, prediction models for Hb deferral have been
developed. Genetic loci have been shown to associate with red cell parameters.
We hypothesize that these SNP*s influence Hb recovery and may have added value
in the prediction models.
Although application of the prediction models may eventually reduce iron
deficiency and decline the number of donor deferrals for low Hb levels, donors
may remain at risk of developing iron deficiency. Therefore, it is valuable to
study the association of blood donation and iron deficiency on consequences of
iron deficiency, such as restless legs syndrome and pica.
Study objective
To investigate the association between genetic factors and Hb recovery after
blood donation. If an association can be found, the added value of genetic
factors in prediction models for Hb deferral will be assessed. Also, the
association between iron deficiency and several health symptoms in blood donors
will be investigated, taking donation frequency into account.
Study design
The design of the study is a prospective cohort study.
In order to investigate the influence of genetic factors on Hb recovery, DNA
profiles of donors with different Hb trajectories will be examined.
Study burden and risks
Participation does not harbour any more risk than ordinary blood donation.

* They must have participated in Donor InSight, and
* The Hb level of their first donor screening must be known,
* Or they must be a new donor who passed the first donor screening
Exclusion criteria
- no written informed consent
- donor does not speak, read and/or write Dutch
- donor is not able to make a decision (Dutch: *wilsbekwaam*)
